An efficient in vitro plant regeneration protocol through callus mediated somatic embryogenesis as well as shoot organogenesis and direct multiple shoot induction has been developed for Pennisetum glaucum. Along with regeneration protocol, a highly efficient Agrobacterium-mediated transformation system has been developed for the production of transgenic pearl millet genotype 843B. Several factors such as optical density of bacteria, co-culture time, co-cultivation duration were optimized for achieving high transformation frequency using shoot tip explants. Standardized Agrobacterium-mediated transformation protocol has been further harnessed for over-expression of AhSIPR10 gene in Pennisetum glaucum. The AhSIPR10 transgenic lines showed increased tolerance to various abiotic stresses.
